Modernized methods for foscarnet

Foscarnet assay and related substances analysis  

Ion chromatography conditions for assay of foscarnet and for determination of impurity B, phosphate and phosphite using Xenoic® XAMS suppressor with ASUREX-A200 automatic regenerator.

Superimposed chromatograms from foscarnet assay calibration acquired on a Shodex IC SI-90 4E column (250x4 mm) using an eluent containing 7.5 mM NaHCO₃ and 7.5 mM Na₂CO₃ in water, pumped at 1.0 mL/min at 25 °C. Background reduced by XAMS suppressor with ASUREX-A200 automatic regenerator. Eluent pumping and conductivity detection by Metrohm 761 Compact IC. Injection of 20 µL of foscarnet (5, 10, 20, 50, 100, and 200 mg/L) in water, giving calibration signals up to about 9.3 µS/cm, or of phosphite (20 mg/L), or of phosphate (5 mg/L), to highlight method specificity.

Ion chromatography separation of foscarnet and related substances on a Metrosep A Supp 16 column (250x4 mm) using an eluent containing 40 mM NaOH and 10 mM Na₂CO₃ in water, pumped at 0.8 mL/min at 25 °C. Background reduced by XAMS suppressor with ASUREX-A200 automatic regenerator. Eluent pumping and conductivity detection by Metrohm 761 Compact IC. Injection of 10 µL of phosphite (10 mg/L), foscarnet impurity B ([ethoxy(hydroxy)phosphoryl] formic acid, 20 mg/L), phosphate (10 mg/L), and foscarnet (phosphonomethanoic acid, 20 mg/L) in water, giving signals up to 2.5 µS/cm.
